福建口岸输入登革I型病毒E基因序列分析

  摘要:目的  研究分析福建口岸输入登革I型病毒E基因序列,为登革热诊断的分子溯源提供参考。方法 收集2012-2013年福建口岸截获的7例入境Ⅰ型登革热病例资料及血清标本,用C6/36细胞培养分离病毒,提取病毒RNA,用RT-nested-PCR扩增包含E基因全长的核酸片段,扩增产物经纯化、测序,用生物学软件进行DNA序列比对分析。结果 用C6/36细胞从7例登革热病例血清中成功分离出7株登革病毒株,经RT-nested-PCR证实均为DENV-Ⅰ型,构建E基因全长进化树分析发现,分离株与新加坡、越南和菲律宾等地的病毒株同源性最高,与病例入境时进行的流行病学调查资料完全吻合。结论 从分子水平证明登革热病例血清中分离到的毒株为DENV-Ⅰ型病毒,结合流行病学调查资料,证实均为输入性感染病例,最有可能来源于东南亚一带。

  Study on E gene sequence of dengue virus type I at Fujianport

  Abstract: Objective  To study E gene sequence of denguevirus type I at Fujian port, and to lay a foundation for moleculardiagnosis of dengue fever. Methods  Epidemiological data andserum samples were collected and viruses were isolated in C6/36cells. The viral RNA was extracted, and amplified to DNA fragmentcontaining the full-length E gene by RT-nested-PCR. The amplifiedproduct was purified, sequenced and analyzed with biologicalsoftware. Results  Seven strains of dengue virus isolates frompatients at Fujian port were shown to be DENV-I. The full length Egene (1485 bp) was amplified successfully. The phylogenetic treeshowed more homology with Singapore, Vietnam and Philippines virusstrains, consistent with the source of entry patients. Conclusion A total of 7 cases were imported dengue fever cases.Southeast Asia was the most likely source.
